转化生长因子-β1在人横纹肌肉瘤肌源性分化中的作用
Role of Transforming Growth Factor-β1 in Myogenic Differentiation of Human Rhabdomyosarcoma

摘要: 目的:探讨转化生长因子-β1(TGF-β1)在横纹肌肉瘤肌源性分化中的作用。方法:采用细胞培养、逆转录多聚酶链式反应(RT-PCR)及免疫荧光染色观察TGF-β1对横纹肌肉瘤细胞系RD肌源性分化相关指标的影响,并用免疫组化SP法,检测49例横纹肌肉瘤组织中TGF-β1表达与肌球蛋白重链(MyHC)之间的相关性。结果:TGF-β1可明显抑制RD细胞肌球蛋白重链mRNA和蛋白的表达。用TGF-β1处理RD细胞后,肌球蛋白重链和肌节肌动蛋白(Sarcomeric-Actin)阳性着色细胞数明显下降(P<0.05)。MyoD1的表达在处理前后未见明显差异。横纹肌肉瘤组织中TGF-β1表达与肌球蛋白重链呈明显负相关(P<0.05)。结论:TGF-β1可抑制RD细胞的肌源性分化,这一作用可能不依赖于MyoD1的调节。TGF-β1信号传导通路可能与横纹肌肉瘤的肌源性分化受阻有关。Abstract: Objective: To evaluate the effect of transforming growth factor-β1 (TGF-β1) in myogenic differentiation of human rhabdomyosarcoma (RMS). Methods: Myogenic differentiation was ana-lyed as the mRNA level of myosin heavy chain (MyHC) by reverse transcription-polymerase chain reaction (RT-PCR). Immunofluorescent staining was used to detect the expression of MyHC, Sarcomeric-Actin and MyoD1. Immunohistochemistry was used to detect the expression of TGF-β1 and MyHC in 49 cases of rhabdomyosarcoma. Results: Exogenous TGF-β1 downregulated expression of MyHC mRNA. Moreover, treatment with TGF-β1 significantly decreased the number of MyHC and Sarcomeric-Actin-expressing cells when compared with the control (P<0.05). However, TGF-β1 treatment didn't change the expression of MyoD1. There was a close negative correlation between the expression of TGF-β1 and MyHC in RMS tissues (r=-0.661, P<0.05). Conclusion: TGF-β1 may inhibit myogenic differentiation independent of its effects on MyoD1 in RD cells. TGF-β1 signal transduction may play an important role in myogenic differentiation of RMS cells.
