The Study on Initiative Immunity of Autogenous Tumor Cell Vaccine after Operation of Esophageal Carcinoma and Cardiac Carcinoma

摘要: 目的:探讨用新城鸡瘟病毒修饰的自体肿瘤细胞疫苗(ATV-NDV)对食管、贲门癌患者临床应用的意义。方法:采用SP一步法和双抗体夹心酶联免疫吸附试验法(ELISA法),对52例接种瘤苗食管、贲门癌手术患者和68例未接种瘤苗食管、贲门癌手术前后的外周血NK活性、T淋巴细胞亚群、白细胞介素-2(IL-2)、白细胞介素-2受体(IL-2R)、白细胞介素-6(IL-6)和肿瘤坏死因子(TNF)进行检测,用40例正常人作对照组。结果:食管、贲门癌手术患者瘤苗接种治疗后细胞免疫指标CD3、CD4百分率和CD4/CD8比值明显增加,CD8百分率明显下降,NK活性无明显变化;食管、贲门癌患者IL-2R、IL-6、TNF水平明显高于正常对照组(P<0.01),而IL-2水平明显较低(P<0.01)。患者术后IL-2R、IL-6较术前明显增高(P<0.01),IL-2、TNF较术前有所降低(P>0.05)。经过免疫接种后,患者IL-2较接种前明显增高(P<0.01),而IL-2R、IL-6、TNF较接种前明显降低(P<0.01)。结论:新城鸡瘟病毒修饰的自体肿瘤细胞疫苗(ATV-NDV)是一种特异性较高的安全有效的免疫制剂,其远期疗效仍在进一步观察中。Abstract: Objective: To evaluate the availability of immune function recovery in postoperative patients with esophageal carcinoma and cardiac carcinoma by Newcastle Disease Virus -modified autologous tumor cell vaccine (ATV-NDV). Methods： The serum level of IL-2、IL-2R、IL-6 and TNF expressions were detected with ELISA from 52 cases of patients with esophageal carcinoma and cardiac carcinoma in preoperation, postoperation and after immunization. Results: In preoperation, the serum level of IL-2R、IL-6 and TNF were higher than the control group, but IL-2 lower than that. In the fourteenth day after surgery the serum levels of IL-2R、IL-6 increased significantly while IL-2、TNF were no significant differences. After immunization, the serum level of IL-2 increased significantly, but IL-2R、IL-6 and TNF decreased significantly than before immunization. There were no distinct adverse effects during treatment. Conclusion： ATV -NDV was a safe, effective and high specially immune pharmaceutics, it remains to be observed in remote effect.
