急性白血病患者MUC1与WT1基因表达的临床意义
Clinical Significance of the Expression of MUC1 and WT1 Gene in Acute Leukemia

摘要: 目的:探讨MUC1基因及WT1基因在急性白血病的表达及临床意义。方法:采用逆转录-聚合酶链反应(RT-PCR)方法,检测54例初发或复发急性白血病(AL)患者MUC1基因以及WT1基因的表达,观察MUC1基因及WT1基因的表达与急性白血病患者疗效关系。结果:54例初治或复发AL中MUC1基因表达阳性率50.0%,WT1基因表达阳性率74.1%。MUC1基因阴性表达的初治非M3型AL19例治疗完全缓解率达94.7%,MUC1阳性表达的初治非M3型AL18例治疗完全缓解率55.6%;两组比较有显著性差异(P<0.05)。WT1基因表达阳性与否与化疗疗效无关。结论:MUC1和WT1基因在急性白血病中有一定的表达,MUC1基因阳性表达可作为急性白血病患者预后不良的一项分子生物学指标。Abstract: Objective: To study the expression of MUC1 and WT1 gene in acute leukemia and their clinical significance. Methods: The expression of MUC1 and WT1 mRNA was detected in 54 newly diagnosed and relapsed patients with acute leukemia by reverse transcriptase polymerase reaction(RT- PCR). With clinical observation, it was analyzed that the relationship of the expression of MUC1 and WT1 gene with the curative effect in the newly diagnosed patients of acute leukemia. Results: The expression of MUC1 was positive in 27 of 54 AL patients (50%) and the expression of WT1 was positive in 40 of 54 AL patients (74.1%). Eighteen of 19 MUC1-negative non-M3 acute leukemia patients(94.7%) got complete remission (CR) while 10 of 18 MUC1-positive non- M3 acute leukemia patients (55.6%) got C R after chemotherapy which showed significance difference (P<0.05=; There are no any relationship between positive expression of WT1 gene and the curative effect in non-M3 acute leukemia. The positive expression of MUC1 and WT1 could turn negative after the patients got CR.Conclusion: The positive expression of MUC1 and WT1 is observed in some acute leukemia patients.MUC1 positive expression may be a bad prognosis factor in acute leukemia.
