水通道蛋白1及4在喉癌组织中的分布及意义

The Expression and Distribution of Aquaporin 1 and Aquaporin 4 in Laryngeal Carcinoma and Its Significance

  • 摘要: 目的:明确水通道蛋白1(aquaporin1,AQP1)及水通道蛋白4(aquaporin4,AQP4)在喉癌组织中的表达和分布,并探讨其在喉癌发病中的意义。方法:取喉癌组织20例及癌旁正常组织15例,应用RT-PCR、Westernblot、免疫组织化学技术检测AQP1及AQP4在喉癌和正常对照组织中的表达及分布。结果:AQP1主要表达于正常喉粘膜固有层的血管内皮细胞及喉粘膜腺体。大量表达于喉癌组织中的血管内皮细胞及部分肿瘤上皮细胞和癌巢中。AQP4主要表达于正常喉粘膜鳞状上皮、假复层柱状纤毛上皮及固有层粘膜腺体,表达于喉癌组织的肿瘤上皮细胞、癌巢及喉粘膜腺体上。结论:AQP1及AQP4分布于喉癌组织的不同部位,协同增加肿瘤上皮和血管对水的通透性运输,从而促进肿瘤的生长及向周围基质浸润。

     

    Abstract: Objective: To confirm the expression and distribution of aquaporin 1 (AQP1) andaquaporin 4 (AQP4) in laryngeal carcinoma and to probe into its significance in the development of la-ryngeal carcinoma.Methods: The tissues in a total of 20 cases of laryngeal carcinoma and paraneo-plastic issues of 15 cases were obtained immediately after surgery. The expression and distribution ofAQP 1 and AQP 4 in laryngeal carcinoma were examined by RT-PCR, Western Blot analysis and im-munohistochemical technique.Results: AQP1 expressed on capillary endothelia and laryngeal mucousglands of normal tissues, and expressed mainly on capillary endothelial cells, part of the tumor cellsand cancer nests of laryngeal carcinoma tissues. AQP4 expressed mainly on squamous epithelial, pseu-dostratified ciliated columnar epithelia and laryngeal mucous glands of normal laryngeal tissues and ontumor cell membrane and cancer nests of laryngeal carcinoma tissues.Conclusion: The distribution ofAQP 1 and AQP 4 on laryngeal carcinoma tissues is dramatically different and may increase water os-motic transport cooperatively in tumor epithelium and vessels, thus promoting the development and themetastasis of tumors.
