X射线照射诱导鼻咽癌细胞mdr l和p53基因的表达及临床意义

The Expression and Significance of X-irradiation in the Induction of mdrl and p53 Gene of Nasopharyngeal Carcinoma Cells

  • 摘要: 目的:探讨X射线照射后鼻咽癌CNE-1细胞产生多药耐药的机制。方法:采用RT-PCR和Westernblot方法,检测X射线照射后CNE-1细胞mdr1和p53基因的表达。结果:照射后细胞mdrlmRNA和P-gp表达均增强,p53蛋白的表达也增强,并与P-gp表达呈正相关性(P<0.05)。结论:X射线照射可诱导CNE-1细胞mdr1和p53基因的表达,两者的相互协调作用可能是照射后肿瘤细胞产生多药耐药的重要机制之一。

     

    Abstract: Objective: To explore mechanism of multi-drug resistance in nasopharyngeal carcino-ma (NPC) CNE- 1 cells after X-irradiation. Methods: The expression of multi-drug resistance (mdr 1)gene and p53 gene was tested by RT-PCR and Western blot. Results: Following irradiation, the ex-pression of mdr1 mRNA and P-gp in cells was increased. The expression of p53 protein was also en-hanced and correlated positively with P-gp expression (P<0.05). Conclusions: Irradiation may inducethe expression of mdr1 and p53 in CNE-1 cells; relationship between these two genes maybe play animportant role in multi-drug resistance after NPC irradiation.
