hMAM Detection in Bone-Marrow Micrometastasis of Patients with Breast Cancer and Its Significance

  • 摘要: 目的 :检测乳腺癌骨髓中 hMAM mRNA表达,判断骨髓微转移状况,探讨其与生物学因子及预后的关系。 方法 :采用 RT-PCR 法检测骨髓组织中hMAM mRNA表达; 免疫组化SP法检测乳腺癌组织中ER 、PR 、Cath- D及VEGF的表达。 结果 :102例Ⅰ~Ⅲ 期乳腺癌患者骨髓组织hMAM mRNA表达阳性率37.3% 。ER 、PR阴性及弱阳性的患者骨髓hMAM mRNA表达率高(P<0.001,P<0.05);骨髓hMAM mRNA表达随Cath-D、VEGF蛋白表达增强而呈增高趋势(P<0.05,P<0.05)乳腺癌患者骨髓hMAM阳性者易发生远处转移(P<0.05)。 结论 :乳腺癌骨髓微转移与癌组织中某些生物学因子有一定相关性;骨髓hMAM阳性患者发生远处转移机会高,微转移检测可以作为判断乳腺癌预后的指标之一。

     

    Abstract: Objective :To detect the expression of hMAM mRNA in bone marrow (BM) of patientswith breast cancer and to determine the relationship between micrometastasis in BM and some molecu-lar markers as well as the prognosis of breast cancer. Methods : The expression of hMAM mRNA in BMof patients with breast cancer was detected using RT-PCR. The expressions of ER, PR, Cath-D andVEGF in cancerous tissues were detected by immunohistochemical s-p method. Results :About 37.3%positive expression rate of hMAM mRNA in 102 patients with stage-ⅠandⅢ breast cancer wasfound. The expression of hMAM in BM was much higher in patients with ER(―)and ER(+)than that ofER(++ 、+++)(P<0.001) and of PR (P<0.05). hMAM expression in BM was significantly positive correlat-ed with Cath-D (P<0.05) and VEGF (P<0.05).There was correlation between the expression of hMAMin BM and the distant metastasis in beast cancer (P<0.05). Conclusions : The micrometastasis in BM iscorrelated with some tumor markers.The patients with positive expression of hMAM in BM have morechance of distant metastasis and may have poor prognosis. The detection of micrometastasis may be oneof the targets to predict the prognosis of breast cancer.
