Survivin及其相关基因在BCML-TA299乳腺癌发展中的作用机制研究

The Mechanism of Action in the Survivin and Related Genes for Development of the BCML-TA299 Breast Cancer

  • 摘要: 目的 :探讨Survivin与其相关基因在乳腺癌侵袭、转移之间的关系。 方法 :利用BCML-TA299乳腺癌肺转移模型,将Suvivin(D-8)和Survivin突变体PAd-T34A瘤内直接注射,动态观察肿瘤的大体形态、病理组织形态;并采用免疫组化技术动态研究相关基因的分子生物学特性;用流式细胞术检测各实验组细胞周期的变化。 结果 :Survivin(D-8)与Survivin突变体PAd-T34A分别促进和抑制乳腺癌的生长和转移,两组中Survivin、C-erbB2、CDK4、Caspase3呈现相反的表达趋势,组间比较有统计学差异(P<0.05)。 结论 :Survivin可能在乳腺癌发生发展中起促进作用,并通过多种机制作用于乳腺癌的转移和侵袭。

     

    Abstract: Objective :To investigate the relationship between Survivin, Survivin-related genesand the invasion and metastasis of breast cancer. Methods : The model for pulmonary metastasis of insitu BCML- TA299 breast cancer was used, then the Survivin (D-8) and Survivin mutant PAD- T34Awere directly injected into the tumors.The genes associated with tumor development were studied by I-HC at different time and the cell-cell cycle of each group by FCM. Results :Survivin (D-8) and Sur-vivin mutant PAD-T34A promote or inhibit the growth and metastasis of breast cancer respectively. Theexpression among the Survivin, c-Erb-2, CDK4 and Caspase3 were adverse (P<0.05). Conclusion :The survivin may promote the development of breast cancer and can effect the invasion, metastasis ofbreast cancer by multiple mechanisms.
