雷公藤内酯醇和肿瘤坏死因子α作用Raji细胞内Notch1蛋白表达的研究

The Studies on Triptolide and TNF-α Regulating the Expression of Notch1 in Raji Cells

  • 摘要: 目的 :通过探讨雷公藤内酯醇(Triptolide,TRL)和肿瘤坏死因子(TNF-α)作用Raji细胞内Notch1蛋白表达的变化,进一步明确Notch信号途径在淋巴瘤发病中的作用。 方法 :采用MTT法检测雷公藤内酯醇对Raji细胞增殖的影响,RT-PCR检测Notch1mRNA含量,流式细胞术分析Notch1蛋白表达的变化。 结果 :1)MTT法结果表明雷公藤内酯醇以时间和剂量依赖的方式抑制Raji细胞增殖;2)RT-PCR检测结果表明雷公藤内酯醇抑制Raji细胞内Notch1mRNA含量,呈剂量依赖的方式,而TNF-α可促进细胞内Notch1mRNA的表达,但此表达可部分被雷公藤内酯醇抑制;3)流式细胞术分析表明在Raji细胞内Notch1蛋白平均荧光强度在TNF-α组明显增加,而在TRL组则表达降低与空白对照组比较有极显著差异(P0.01)。 结论 :1)在Raji细胞内,存在Notch1蛋白的表达;2)雷公藤内酯醇可抑制细胞内Notch1蛋白的表达,而TNF-α可上调Raji细胞内Notch1蛋白的表达;3)TNF-α和雷公藤内酯醇可通过影响Notch信号途径在淋巴瘤细胞内发挥作用。

     

    Abstract: Objective : To investigate the effect of TNF-α on the expression of Notch1 for better understanding a novel mechanism for the neogenic capacity of Notch1 in Raji cell lines and elucidation of the effect of activated Notch1 signaling in Raji cell lines. Methods : The effects of triptolide on the proliferation of Raji cells were tested using MTT; the mRNA level of Notch1 in Raji cell line treated by TNF-α and triptolide was measured by RT-PCR. The change of expressions of Notch1 protein was tested by flow cytometry. Results : a) Triptolide inhibited the proliferation of Raji cells in a dose- and time-dependent manner. b) Triptolide reduced the Notch1 mRNA level in a dose-dependent manner compared with the controls in Raji cells. The Notch1 mRNA level increased after treated by TNF-α (P0.01). c) Flow cytometry also showed increased level of Notch1 protein induced by TNF-α, and TRL inhibited the expression of Notch1 protein in Raji cells. Conclusion : Notch1 protein is expressed in Raji cells. TNF-α and triptolide can play a role through affecting Notch1 signaling and that it might be a potential new target for treatment.
