缺氧诱导结直肠癌细胞Ang-2的表达

Hypoxia-induced upregulation of Angiopoietin-2 in Colorectal Cancer

  • 摘要: 目的:探讨缺氧对结直肠癌(colorectalcancer,CRC)细胞表达血管生成素-2(angiopoietin,Ang-2)的诱导作用,将其作为抗肿瘤血管生成的治疗靶点。方法:采用RT-PCR及免疫组化法分别检测3个结直肠癌细胞系及CRC肝转移标本的Ang-2及葡萄糖转运蛋白1(glucosetransporter1,GLUT-1)的表达,用CD31单克隆抗体标记血管内皮细胞并计数微血管密度(microvesseldensity,MVD)。结果:HCT-116细胞系氧供正常时不表达Ang-2,缺氧可诱导其表达;LoVo细胞系仅在低血清缺氧培养48h可检出Ang-2mRNA表达;HT-29细胞系在氧供正常及缺氧情况下均可检出Ang-2mRNA,二者无显著差异。肝转移癌中心Ang-2表达水平最高,而MVD情况正相反。GLUT-1的表达模式与Ang-2相似。结论:缺氧在诱导CRC表达Ang-2中具有重要作用,低营养等其它因素与Ang-2的表达可能有关。

     

    Abstract: Objective: To determine the role of hypoxia in stimulating the expression of angiopoietin-2 (Ang-2) in colorectal cancer (CRC). Methods: The expression of Ang-2 and glucose transporter-1 (GLUT-1) were examined in 3 cultured CRC cell lines by reverse transcriptase-polymerase chain reaction (RT-PCR) assays and in specimens of hepatic CRC metastasis using immunohistochemistry (IHC). Results: Hypoxia-induced Ang-2 mRNA expression was observed in the HCT116 cell line that did not express Ang-2 under normoxic conditions. In the LoVo cell line, the Ang-2 mRNA was only detectable at 48 hr in hypoxic serum-deprived cultures. In the HT29 cell line, Ang-2 mRNA could be detected under both normal oxygen supply and anoxic conditions, without noticeable difference. A stepwise increase of Ang-2 expression from the periphery to the central part of liver metastatic foci was noted whereas vascular density had the reverse pattern. An expression pattern similar to that of Ang-2 was found in GLUT-1, a known hypoxia-induced factor. Conclusion: The present study suggests that hypoxia may play an important role in inducing the expression of Ang-2 in CRC.
