摘要: 目的:观察不同浓度ω-3多不饱和脂肪酸(EPA、DHA)对人胃癌BGC-823细胞存活情况、PGE2和脂质过氧化的影响。方法:采用MTT法测定肿瘤细胞存活率,酶联免疫吸附竞争法测定PGE2浓度,黄嘌呤氧化酶法测定总SOD活力,TBA法测定MDA含量。结果:30μg/ml和45μg/ml的EPA或DHA可显著降低肿瘤细胞存活率(P<0.001),细胞PGE2浓度和总SOD活力明显下降(P<0.05)、而MDA含量明显升高(P<0.05)。结论:ω-3多不饱和脂肪酸可通过PGE2代谢和脂质过氧化作用抑制人胃癌BGC-823细胞的生长。Abstract: Objective: To observe the effect of different concentrations of omega-3 polyunsaturated fatty acids on cell viability, prostaglandin E2 (PGE2) production and lipid peroxidation in the human gastric cell line BGC-823. Methods: The survival rate of tumor cells was determined by MTT assay, and PGE2 level was established with competitive ELISA. Total superoxide dismutase (SOD) was examined with the xanthine oxidase test and malondialdehyde (MDA) was detected using thibabituric acid (TBA). Results: After treatment with 30μg/ml or 45μg/ml of eicosapentaenoic acid (EPA) or docosahexaenoic acid (DHA), the survival rate of tumor cells was markedly decreased (P<0.001). The concentration of PGE2 and total SOD in the treated cells was decreased (P<0.05), whereas MDA was increased (P<0.05). Conclusion: Omega-3 polyunsaturated fatty acids can inhibit the growth of cultured cells through a mechanism involving PGE2 metabolism and lipid peroxidation.
