  • 摘要: 目的:研究胃癌组织中Survivin基因表达的mRNA拷贝数、细胞中蛋白表达及凋亡情况,结合临床资料分析三者间及其与病理的关系,讨论其在胃癌诊断、预后等方面的意义。方法:SurvivinmRNA拷贝数采用分子信标荧光定量PCR方法,以其读码框部分cDNA克隆重组子为标准品进行绝对定量测定;细胞中蛋白表达采用免疫组化方法;细胞凋亡采用TUNEL方法。结果:SurvivinmRNA拷贝数和蛋白表达阳性率,肿瘤组织显著高于正常组织(P<0.05);细胞凋亡指数AI,肿瘤组织显著低于正常组织。肿瘤组织中SurvivinmRNA拷贝数与患者的生存状况和病理类型显著相关,且与AI呈显著负相关(r=-0.252,P<0.05);Survivin蛋白表达阳性率与患者生存状况显著相关,蛋白表达阳性组mRNA拷贝数高于蛋白表达阴性组,AI低于表达阴性组,但无统计学意义(P>0.05);SurvivinmRNA表达和蛋白表达无显著相关性(P>0.05)。结论:SurvivinmRNA和蛋白在胃癌中表达上调,可作为判断预后的生物学指标。

     

    Abstract: Objective: To study the mRNA copy number of survivin, protein expression of survivin and the apoptotic index (AI) in gastric carcinomas, to analyze the relationships between these 3 clinical parameters and pathologic results and to discuss the significance in terms of diagnosis and prognosis. Methods: The copy number of survivin mRNA was measured by quantitative PCR with a molecular beacon probe, using a cDNA clone as a standard for the absolute quantitative assay. The protein expression was investigated by immunohistochemistry (IHC) SP method, and the apoptotic index was analyzed with TUNEL. Results: The copy number of survivin mRNA and positive rate of protein expression were higher in tumor tissues than in the normal controls (P<0.05) and the apoptotic index was lower in tumor tissues than in normal tissues. There was a significant positive correlation between the copy number of survivin mRNA and the survival time and pathological types, and there was a negative correlation between the copy number of survivin mRNA and AI (r=-0.252, P<0.05). There was a significant correlation between the positive rate of survivin protein expression and survival time. The copy number of mRNA in the positive protein expression group was higher than in the negative expression group, and the AI was lower in the positive expression group than in the negative expression group, but there was no statistical significance between them (P>0.05). There was no significant correlation between the expression of survivin mRNA and protein (P>0.05). Conclusion: The copy number of survivin mRNA and protein expression rates were higher in gastric carcinoma than in normal controls. Survivin mRNA and protein levels may be potential molecular indicators for prognosis. There is upregulation of survivin mRNA and protein expression in gastric cancer, which can be used as biological index for diagnosis.
