卵巢癌组织中clusterin蛋白表达和细胞凋亡检测

Clusterin Protein Expression and Apoptosis in Ovarian Cancer

  • 摘要: 目的:探讨clusterin蛋白在卵巢癌中的表达及其与细胞凋亡的关系和临床意义。方法:制作86例卵巢癌的组织芯片,分别应用免疫组织化学和TUNEL方法,检测卵巢癌中clusterin蛋白表达及细胞凋亡情况,结合临床病理学资料,分析其临床意义。结果:在组织芯片中,73例卵巢癌成功进行了免疫组化和TUNEL染色,其中38例(52%)出现clusterin蛋白的过度表达,在不同临床分期的卵巢癌中,clusterin蛋白表达有显著性差异(P<0.05),clus-terin蛋白过度表达多见于临床晚期(Ⅱ期及以上)的卵巢癌。另外,clusterin过度表达的肿瘤多呈现低的细胞凋亡指数(apoptoticindex,AI),而clusterin正常表达的肿瘤,则大多数表现为高AI值(P<0.05)。结论:clusterin蛋白在卵巢癌中过度表达与肿瘤的临床分期密切正相关,与肿瘤细胞凋亡显著负相关。

     

    Abstract: Objective: To investigate the clinical significance of clusterin protein expression in ovarian cancer and its association with tumor cell apoptosis. Methods: A tissue microarray (TMA) containing 86 cases of epithelial ovarian carcinoma was constructed. Immunohistochemistry (IHC) was used to examine clusterin protein expression and TUNEL staining was used to determine the apoptosis index.These data were used to determine the clinical significance in relation to clinicopathological data. Results: Using the ovarian cancer TMA, IHC and TUNEL staining were successfully conducted in 73 cases of ovarian cancer. Overexpression of clusterin protein was observed in 38 cases (52%). There was a significant difference in clusterin expression between tumors of different clinical stages (P<0.05) where the overexpression of clusterin was frequently seen in advanced stage tumors (Ⅱ-Ⅳ). In addition, a lower apoptosis index (AI) was observed in the epithelial ovarian carcinomas with overexpression of clusterin, while a higher AI was shown in most of the tumors with normal clusterin expression (P<0.05).Conclusions: Overexpression of clusterin protein in ovarian cancer is positively correlated with the tumor's clinical stage and is negatively correlated with tumor cell apoptosis.
