纳米控释紫杉醇对小鼠乳癌的凋亡诱导研究

摘要: 目的:研究纳米紫杉醇体内对BCML-TA299乳癌的抑制作用和凋亡的诱导。方法:建立小鼠体内荷BCML-TA299乳癌模型,用纳米紫杉醇载体注入小鼠皮下,观察肿瘤生长情况并计算肿瘤体内抑瘤率,应用HE染色形态学观察、BaxmRNA原位杂交以及流式细胞仪检测等方法,了解纳米控释紫杉醇诱导小鼠乳癌细胞凋亡的作用。结果:纳米紫杉醇10mg/kg、15mg/kg给小鼠乳癌原位注射,对BCML-TA299乳癌有明显抑制作用和诱导凋亡作用,且有剂量依赖关系。结论:纳米紫杉醇体内对BCML-TA299乳癌模型细胞增殖均有抑制作用,诱导肿瘤细胞死亡的主要途径可能是凋亡。Abstract: Objective: To study the inhibitory action and induced apoptotic effect of Paclitaxelnanometer (NP) on BCML-TA299-bearing mice in vivo. Methods: The inhibitory effect on tumors was observed in BCML-TA29-bearing mice. The expression of bax mRNA was assessed by in situ hybridization and flow cytometry. Growth inhibition was assessed and microscopy was also utilized. The data were used to examine the effect of Paclitaxel-NP on BCML-TA299 breast cancer. Results: The growth of BCML-TA299 was markedly inhibited by Paclitaxel-NP in vivo in a concentration-dependent manner. The expression of proapoptotic Bax was increased. Conclusion: Paclitaxel-NP may significantly inhibit the growth of breast cancer in mice by inducing apoptosis and inhibiting proliferation of the tumor cells.
