Inactivation of the RUNX3 Gene and Gastric Cancer Carcinogenesis

摘要: RUNX3基因是新近发现的一种抑癌基因,属人runt相关转录因子(human runt-related transcription fac-tor,RUNX)家族成员,是TGF-β信号传导通路中的一个重要环节,杂合子丢失(LOH)和启动子高甲基化可使其失活,并可导致胃癌的发生、发展和转移。本文综述了RUNX3基因的结构、功能及其与胃粘膜异常增生与癌变的关系。Abstract: As one of the human runt-related transcription factors, the RUNX3 gene is found to be an anti-oncogene. It plays an important role in the TGF- βsignaling pathway. The mechanism of RUNX3 inactivation involves loss of heterozygosity (LOH) and promoter hypermethylation, resulting in the development and metastasis of gastric cancer. The structure and function of RUNX3 and the relationship between the RUNX3 gene and gastric mucous hyperplasia as well as gastric cancer are reviewed in the article.
