PTEN基因突变及mRNA表达与卵巢癌病理生物学特征的关系

Relationship between the Mutation and mRNA Expression of PTEN Gene and Pathobiological Behavior of Ovarian Cancer

  • 摘要: 目的:研究抑癌基因PTEN突变和mRNA表达在卵巢癌发生和演进中的作用.方法:利用PCR-SSCP方法检测正常卵巢(n=5)、上皮性卵巢囊肿(n=5)、卵巢交界性肿瘤(n=9)、卵巢癌(n=60)中PTEN外显子5的突变,采用RT-PCR检测这些组织中mRNA表达,比较PTEN基因突变和mRNA表达与卵巢癌临床病理特征的关系.结果:仅在上皮来源的卵巢癌中发现5例(7.1%)PTEN基因外显子5突变,PTEN基因mRNA在卵巢交界性肿瘤和卵巢癌中的表达水平低于正常卵巢和卵巢囊肿(P<0.05),PTEN基因mRNA表达与卵巢癌的临床病理分期呈负相关(P<0.05),而与组织分化程度呈正相关(P<0.05),PTEN基因mRNA表达在卵巢内膜样癌中表达最低,显著低于浆液性或粘液性卵巢囊腺癌(P<0.05).结论:卵巢癌中存在PTEN基因外显子5突变及mRNA表达下调.PTEN基因mRNA表达下调与卵巢上皮性恶性肿瘤,特别是内膜样腺癌的发生和病理生物学行为关系密切.因此,PTEN基因有望作为反映卵巢癌病理生物学行为的客观分子指标.

     

    Abstract: Objective: To investigate the role of mutation and mRNA expression of tumor suppressor gene PTEN in tumorigenesis and progression of ovarian cancer. Methods: Mutated exon 5 of PTENgene was detected in normal ovary (n=5), epithelia ovarian cyst (n=5), epithelia ovarian borderline tumor (n=9), ovarian cancer (n=60) by PCR-SSCP. mRNA expression of PTEN gene was evaluated in cor-responding tissues by RT-PCR. The mutation or mRNA expression of PTEN gene was compared with clinicopathological features of ovarian cancer. Results: Mutated exon 5 of PTEN gene was examined only in 5 (7.1%) cases of epithelium-derived ovarian cancer. mRNA expression level of PTEN gene was lower in ovarian borderline tumor and ovarian cancer than that in normal ovary and ovarian cyst (P <0.05). mRNA expression level of PTEN gene was negatively correlated with clinicopathological staging,but positively with histological differentiation (P < 0.05). mRNA expression level of PTEN gene was thelowest in ovarian endometrioid cancer, significantly lower than ovarian serous and mucinous cancer (P <0.05). Conclusions: Mutated exon 5 and reduced mRNA expression of PTEN gene occurred in ovariancancer. Down -regulated mRNA expression of PTEN gene was closely associated with pathobiological behavior of epithelial ovarian cancer, specially of ovarian endometrioid cancer. Thus, PTEN gene might be a pathobiological molecular marker oi ovarian cancer.
