RKIP与NF-κB在肺癌侵袭转移中的作用

Role of Raf Kinase Inhibitor Protein and Nuclear Factor-κB in the Invasion and Metastasis of Lung Carcinoma

  • 摘要: 目的:检测肺癌组织中Raf激酶抑制蛋白(raf kinase inhibitor protein ,RKIP)和核因子κ B(nuclear factor-κ B,NF-κ B)的表达,分析其与临床病理特征及预后的关系。方法:应用免疫组织化学方法(SP法)检测83例肺癌及65例癌旁正常肺组织中RKIP与NF-κ B 蛋白的表达情况。结果:83例肺癌中,RKIP蛋白的低表达率及NF-κ B 蛋白的高表达率分别为20.5%(17/83)、68.7%(57/83),其表达水平与肺癌的分化程度、TNM分期、淋巴结或远处转移及术后生存时间有明显关系(P 均<0.05);RKIP蛋白的表达水平与NF-κ B 呈负相关(P<0.05);RKIP蛋白低表达与NF-κ B 蛋白高表达的病例平均生存时间和5 年生存率明显降低(P<0.05)。 结论:RKIP低表达与肺癌的侵袭、转移密切相关,RKIP可能是肺癌的一个转移抑制蛋白,RKIP表达下调可能通过活化NF-κB 信号通路促进肺癌细胞的侵袭和转移。
    Abstract: Objective:To investigate the expression of raf kinase inhibi tor protein (RKIP) and NF-κ B in lung carcinoma and its correlation with clinicopathologic features and survival time. Methods:RKIP and NF-κ B protein levels were detected using immunohistochemistry in 83lung carcinoma specimens and 65non-tumor lung samples.Results: Immunohistochem-istry revealed that of the 83carcinoma cases, there was low expression of RKIP and high expression of NF-κ B protein, 20.5% (17/83) and 68.7% (57/83), respectively. Expression levels of RKIP and NF-κ B protein were correlated wi th tumor differentiation, TNM staging, lymph node metastasis, and survival time (P<0.05, P<0.05, P<0.05, P<0.05, respectively). RKIP expression was negatively correlated wi th NF-κ B expression in lung carcinoma (P<0.05). The mean survival time in cases with a low expression level of RKIP protein was significantly shorter than those with a high expression level, whereas the mean survival time in cases wi th a high expression level of NF-κ B protein was signi ficantly shorter than those wi th a low expression level ( P<0.05). Conclusion: Expression of RKIP and NF-κ B are closely correlated wi th invasion and metastasis of lung carcinoma. RKIP may be a lung carcinoma metastasis suppressor, and decreased RKIP expression is associated wi th the increased metastatic potential of lung carcinoma cel ls through the activation of the NF-κB signal ing pathway.
